In this paper, the author focuses on the process of gene cloning, particularly the cloning a TROVE2 Gene to a PNEB193 Plasmid. This paper answers the research question on whether it is possible to clone TROVE2 Genes to PNEB193 Plasmid. Cloning is a complicated process that involves the combination of various significant molecular processes, both natural and artificial; in general, copies of a specific gene can be generated through cloning.
The sheer complexity of the concepts and principles surrounding cloning appears to be the major reason why not a lot of researchers delve into the topic and why not a lot of researches that are aimed at answering a research question or two about both general and specific cloning mechanisms have been published. In this study, the researchers attempted to clone the gene TROVE 2 after it has been obtained from the K562 cell line ©DNA, and then transforming it into a PNEB193 plasmid.
In order to clone the gene successfully, the researchers had to precisely follow a series of steps that started from the linearization and purification of phosphate-treated plasmid and ended with the isolation of recombinant plasmid from transformed E-coli cells (specific steps were listed in the methodology section).
The key area to discuss here would be whether the researchers were actually able to clone their target gene into their target organism (i.e. E-coli cells). Following all the steps, the researchers were able to successfully insert the TROVE2 gene in the E-coli sample microorganisms—specifically white and blue E-coli colonies, suggesting that the cloning procedure was a success. This was evidenced by the observations made in the plasmid map that was generated as shown in the figures that were presented in the study.
Overall, the entire study was a success. However, further research and analysis are needed in order to create more generalizable outcomes and results. This is because this particular journal was limited to the application of the proposed methods on a limited number of microbes only. Additionally, there was only a single cloning verification method (to measure the success of the procedure) that was used (i.e. use of band detection on plasmid maps). Now, people, particularly the academic community, should care about the outcome of this study because it sets the stage for further research on the possibility of cloning genes of more sophisticated organisms.
