Induction of Apoptotic Program in Cell-free Extracts: Requirement for dATP and Cytochrome c
This research sought to establish that cells that are through the process of apoptosis would have an increase the level of cytochrome c to their existing cytosol during vivo. This would infer that mitochondria of the cells would continue to work during apoptosis activity thereby initiating the production of cytochrome c The apoptotic program is brought about by the introduction of dATP. According to this research the cytosol of the cell produced approximately 15 kDa protein. This amount of protein meets the threshold that can initiate vitro apoptosis. Based on the protein patterns it was clear that the protein produced in this case was cytochrome c. When the produced cytochrome c in cytosol of the cell is removed, it was observed that the level of apoptotic activity significantly reduced. When cytochrome c was reintroduced to the initial extract the level of apoptotic activity increased.
